David Sinclair Bio, Life, and Career:

David Andrew Sinclair AO, an Australian scientist who teaches genetics and serves as co-director of the Paul F. Glenn Center for Biology of Aging Research at Harvard Medical School, was born on June 26, 1969. Sinclair found that sirtuin reduces ageing in yeast by lowering the buildup of extrachromosomal rDNA circles when he was working at Guarente’s lab. NAD was recognized at the time by other lab workers as a crucial cofactor for sirtuin activity.

He disagreed with Guarente at a scientific gathering at Cold Spring Harbor Laboratory in 2002, after he had departed for Harvard, and challenged Guarante’s explanation of how sir2 may be implicated in aging. This sparked a scientific rivalry. Sinclair has won multiple honors for his work, including the Australian Society for Medical Research Medal in 2014, the Advance Award in Life Sciences from the Australian government, and the Irving S. Wright Award of Distinction from the American Federation for Aging Research in 2018.

Sinclair was named one of the 100 most important persons in the world by Time magazine in 2014, and he was named one of the 50 Most Influential People in Healthcare by Time magazine in 2018.

For “distinguished contribution to medical research into the biology of aging and lifetime extension, as a geneticist and academic, to biosecurity efforts, and as an advocate for the study of science,” Sinclair was named an officer of the Order of Australia in 2018.

When Sinclair’s lab was still tiny, in 2003, researchers at the Pennsylvania biotech business Biomol Research Laboratories had found that polyphenols, such as resveratrol, might activate sir2. Sinclair worked with them to validate this discovery.

As a result, Sinclair contributed to articles that were published in Science and Nature in 2003. A scientific debate over whether this was accurate and if resveratrol really activated sirtuins was sparked by Sinclair’s vocal support of resveratrol as an anti-aging substance.

Intense criticism has also been directed at prominent articles that claimed that aging could be reversed in mice.  Increases in the number of mitochondria were discovered in the cells of mice given large doses of resveratrol, according to research from another lab that was partly funded by Sirtris.  In order to understand aging and how to delay it, Sinclair’s group continues to study resveratrol and its analogs, mitochondria, and NAD. According to him, developments in the study of aging might make it possible for people to live to be 200 years old.

He is renowned for his work on epigenetics-focused aging studies. He is a member of the Australian Order. When Leonard P. Guarente, a professor at the Massachusetts Institute of Technology who researched genes implicated in the control of aging, visited Australia in 1993 for a lecture tour, Sinclair was inspired to seek for a post-doctoral job in Guarente’s group.

A single gene mutation in had been identified earlier that year by Cynthia Kenyon’s team at UCSF as having the ability to increase the lifetime of C. elegans. involvement with the company and its marketing, Sinclair objected to the use of his name and words to promote the supplement and left the board. Based on research conducted by Tilly on mammalian oogonial stem cells and work by Sinclair on mitochondria, Sinclair co-founded OvaScience in 2011 alongside Michelle Dipp, Aldrich, Westphal, and others.

Tilly’s research generated controversy since other organizations were unable to duplicate it. As a result of avoiding US regulatory authorities for fertility testing, the firm finally came under fire. Sinclair, together with Nir Barzilai and other associates, co-founded CohBar in 2011. The goal of CohBar was to identify and create brand-new peptides generated from mitochondria. Despite not having any medication candidates during clinical development, Cohbar touts itself as a clinical-stage biotechnology business.

When seeking financing for his lab, Sinclair explained to The Scientist in 2015 how his lab expanded to around 20 employees, dropped back down to about 5, and then grew once again when he received money from charitable groups and businesses, including businesses he helped build.

His lab employed 22 individuals in 2015, had one R01 award and received 75% of its funding from non-federal sources. This was no longer the case as of 2016 when his governmental funds started to rise.

This was also made available on Audible as an audiobook, read by Sinclair.  Sinclair extensively describes his methods for extending life on social media and incorporates them into the book. They include NMN and resveratrol dosages taken every day, which Sinclair describes as SIRT1 activators. The removal of NMN off the market as a supplement by Sinclair’s business Metro Biotech in November 2022 drew criticism since Sinclair had both registered NMN and made it known that it was an experimental novel medicine.

After serving as Guarente’s postdoctoral researcher for four years, Sinclair was employed by Harvard Medical School in 1999. His lab was modest and had financial difficulties in 2003.  Paul F. Glenn gave $5 million to Harvard to establish the Paul F. Glenn Laboratories for the Biological Mechanisms of Aging, of which Sinclair was named the inaugural director. Sinclair and Glenn first met in 2004. Together with Bruce Yankner, is the co-director at the moment.

Sinclair created Sirtris Pharmaceuticals alongside serial entrepreneur Andrew Perlman, Christoph Westphal, Richard Aldrich, Richard Pops, and Paul Schimmel in 2004. Sirtris was primarily concerned in expanding Sinclair’s investigation into sirtuin activators, which was started at the Guarente lab.

Sinclair gained notoriety for describing resveratrol as ” as close to a miraculous molecule you can find… One hundred years from now, people may be taking these molecules on a daily basis to prevent heart disease, stroke, and cancer.” The company was specifically focused on resveratrol formulations and derivatives as activators of the SIRT1 enzyme.

The majority of the anti-aging sector was more circumspect, particularly in light of resveratrol’s low bioavailability and potential additional physiological effects. The company’s first offering was a resveratrol compound known as SRT501. Sirtris went public in 2007, and GlaxoSmithKline later paid $720 million to acquire it and turn it into a subsidiary. Five years later, GSK ended the Sirtris program without developing a viable medication.

Sinclair was a co-founder of Genocea Biosciences, a company created in 2006 on the research of Harvard scientist Darren E. Higgins about antigens that excite T cells and the application of these antigens to make vaccines. Genocea terminated the majority of its employees in 2022 after providing unsatisfactory data at AACR. At Harvard Medical School, Sinclair received a promotion to tenured professor in 2008.

A few years later, he was also appointed as a joint professor at the University of New South Wales School of Medical Sciences. Time, The New York Times, The Charlie Rose Show, 60 Minutes, Boston, The Washington Post, The Economist, TED, and The Joe Rogan Experience have all featured Sinclair. David Andrew Sinclair was raised in St Ives, New South Wales, and was born in Australia in 1969.

Following the 1956 repression of the Hungarian Uprising, his paternal grandmother immigrated to Australia, and his father changed the family name from Szigeti to Sinclair. With an emphasis on yeast gene control, Sinclair completed his studies at the University of New South Wales, Sydney, earning a BSc in biochemistry with honors in 1991 and a Ph.D. in molecular genetics in 1995. He also received the Commonwealth Prize for Australia.
